What Is Mutation Breeding?

Mutation breeding refers to a breeding method that uses physical, chemical and other factors to induce mutations in plant genetic material, thereby developing new crop varieties with target traits. Mutation breeding plays an important role in the development of superior plant varieties and has been widely used to improve crop traits. The spontaneous mutation rate of plants is low, and a variety of physical, chemical, and biological mutagenesis methods can be applied to increase their mutation frequency.

What We Offer

Plant mutations can be induced by physical stimulations (e.g., X-rays, α- and β-particles, fast neutrons, and UV light), which often lead to chromosome changes, or by chemical treatments (e.g., ethyl methanesulfonate (EMS)) which typically cause point mutations. Alternatively, mutations can also be introduced through advanced genetic engineering technologies such as CRISPR/CAS9 and RNAi.

Mutations not only can be easily detected by regular polymerase chain reaction (PCR) approaches, but also can be screened by high-throughput technologies such as whole genome sequencing.

TILLING

Targeting induced local lesions in genomes (TILLING) is also a popular method that allows high-throughput detection of random mutagenesis within the genome. TILLING is a highly feasible technology, which has been applied in a variety of plant species including Arabidopsis thaliana, Medicago truncatula, Lotus japonicus, Brassica rapa, tomato,rice, maize, barley, lotus, poplar, and wheat.

Different types of TILLING

  • Deletion-TILLING: a TILLING variant based on a neutron-induced mutagenesis system called Deleteagene (Delete-a-gene).
  • High Resolution Melting (HRM) - Based TILLING: a high-throughput method that relies on DNA melting curves derived from fluorescent dyes.
  • Conformation-Sensitive Capillary Electrophoresis (CSCE)-Based TILLING: a method for which detection of mutations is based on heteroduplex analysis of DNAs.
  • Eco-TILLING (ecotype TILLING): an efficient method for detecting SNPs in nature population.
  • DEco-TILLING: a cheaper and easier modification of TILLING and Eco-TILLING.
