Extraction Methods of Endoplasmic Reticulum in Plants

  • To extract endoplasmic reticulum from leaves by sucrose density gradient centrifugation, it is necessary to prepare plant leaf homogenate, prepare sucrose density gradient solution, and finally obtain bands with different colors and widths. Endoplasmic reticulum is mainly enriched in the light brown liquid.
  • Endoplasmic reticulum is extracted by differential centrifugation combined with calcium chloride precipitation. After adding CaCl2 solution to plant leaf homogenate, the fractions of rough endoplasmic reticulum (ER) are precipitated.
  • Endoplasmic reticulum is extracted by differential centrifugation, including overspeed, and the precipitate obtained is the total ER fraction by centrifugation at different speeds for different times.
