Application of Exosomes in Facial Beauty

Extracellular vesicles (EVs) are lipid bilayer vesicles released into the extracellular space from nearly all cell types. EVs are divided into three types based on their size and biogenesis: exosomes, microvesicles (MVs), and apoptotic bodies.

Exosomes could be an ideal alternative to other nanocarriers for improving skin conditions. They naturally carry a variety of biological molecules such as proteins, lipids, DNA and a range of RNAs that are critical for intercellular communication and modulate the molecular activity of receptor cells.

In addition, they are well tolerated in the body, readily taken up by cells, and can be targeted for uptake by specific tissues.

Application of exosomes in facial beauty

Anti-wrinkles, anti-aging and anti-pigmentation are the facial beauty features most commonly sought by consumers. Exosomes, a new class of nanocarriers with various advantages, have been introduced to enhance these effects in cosmetics.

Reduce scar formation

Surgery, laser treatments, chemical peels, dermabrasion, steroid or collagen injections and ointments can minimize or eliminate scarring. Exosomes can accelerate wound healing and promote scar-free healing through reorganized collagen fibers that are less cross-linked and have a flatter epidermal surface. EVs can promote skin cell proliferation, migration and angiogenesis.

Anti-aging

A variety of anti-aging options already exist to improve the appearance of aging skin, namely cosmetics, phototherapy and microneedling. However, the improvements observed did not last long. Exosome therapy exerts anti-aging effects by enhancing fibroblast activity, regulating collagen turnover, reducing ROS production and reducing tissue inflammation, and may become a better choice for anti-aging.

Exosomes can promote the migration and proliferation of fibroblasts. Exosome therapy prevents fibroblasts from cycle arrest and reduces the expression of cell cycle arrest-related proteins P53 and P21. Human iPSC-derived exosomes (iPSC-Exo) eliminate senescence-associated β-galactosidase (SA-β-Gal) production. Exosome treatment also protects fibroblasts from UVB-induced senescence. These findings suggest that exosome treatment can prevent fibroblast senescence and restore the regenerative capacity of aging fibroblasts.

Regulate pigmentation

Exosome treatment can slow down UVB-stimulated melanin production and promote melanosome degradation. Keratinocyte and amniotic stem cell-derived exosomes reduce cellular melanin content by reducing key melanogenesis-related proteins (TYR, TYRP1, and TYRP2) and MITF, which regulates these proteins. In addition, exosomes can also promote melanosome degradation by activating autophagy in the mouse melanoma cell line of C57BL/6J mice. The authors attributed the anti-pigmentation effect of exosomes to the contents of miR-330-5p, miR-181a-5p and miR-199a. Furthermore, UV exposure induces melanocytes to produce exosomes, and melanocyte-derived exosomes induce anti-apoptotic signaling and enhance keratinocyte proliferation and migration. These results indicate that exosomes can reduce skin pigmentation under normal conditions and act as paracrine factors to mitigate damage caused by UV exposure.

Overall, exosomes can be used to promote anti-aging, anti-pigmentation and scar-free wound healing, and they can be combined with other technologies such as microneedling for better results.
