Traditional methods of phage DNA extraction are cumbersome and time-consuming and require specialized equipment such as ultracentrifuges.

Services content

The following is the specific procedure for agricultural phage DNA extraction:

  • Separate and purify the phage suspended in ribonuclease solution and incubate at 37 °C in a shaker for a certain length.
  • Transfer the liquid to a centrifuge tube, add the SDS mixture and leave it in a hot water bath for a certain period.
  • Immediately after removal, add potassium acetate and ice bath.
  • Centrifuge with a micro high-speed centrifuge, transfer the supernatant to a new tube, mix with an equal volume of phenol/chloroform, and shake manually.
  • Centrifuge with a micro high-speed centrifuge, separate the two phases, transfer the supernatant to another new tube and add isopropanol.
  • Centrifuge with a micro high-speed centrifuge to precipitate DNA, add a buffer to dissolve DNA, and store at -20 °C.

Things to note for customers

  • When the phage and host bacteria used for lysis are obtained in a fresh culture, lysis is good, and lysis phage harvest is enormous.
  • When liquid culture lysis, if lysis has not occurred by the time of incubation, the incubation temperature can be appropriately increased, or the shaking speed can be increased.
  • When phenol/chloroform extraction, pay attention to the contamination of impurities such as PEG and protein, which can affect restriction endonuclease cleavage.
