Construction of the brazzein expression vectors Vectors such as pPpT4_Alpha_S can be used to express brazzein. The gene sequence of brazzein is amplified, a suitable restriction site is selected, and the brazzein gene is cloned into the vector. If necessary, we will also optimize codons according to the target strain and plasmid.
Transformation and optimization of culture conditions Transfer brazzein into the target strain by plasmid-mediated or electrotransformation. Then we optimize the culture conditions of the strain through a series of measures, such as carbon source, nitrogen source, and other nutrients, etc., to finally improve the expression of brazzein.
Analysis of recombinant brazzein Purify the recombinant brazzein expressed by the microbial strain and then analyze and verify it by WB and mass spectrometry. We are also able to analyze the function of recombinant brazzein using human embryonic kidney cells (HEK293T) stably transfected with chimeric Gα16gust44 protein.
