Microbial strain selection In addition to customer-determined strains, we can also choose microbial strains according to your purpose. Bacteria and fungi are all optional content for us.
Construction and amplification of neoculin expression plasmid Select the appropriate restriction site according to the gene sequence, clone the neoculin gene, and connect the gene to the expression plasmid. The constructed plasmid is transferred to E.coli DH5α for amplification.
Transformation of strains Using the transformation test, the plasmid expressing neoculin is transferred into the target strain. Transformants are then screened for neoculin production.
We will also optimize temperature, medium, trace elements, etc., and explore the culture conditions conducive to large-scale production of neoculin.
Purification of recombinant neoculin We precipitate neoculin in a liquid medium, then purify and collect neoculin by dialysis, filtration, and chromatography for further analysis.
Neoculin characterization We will prepare anti-neoculin antibody to detect both recombinant neoculin and natural neoculin.
Tests of sweetness response and sensory analysis We culture human embryonic kidney cells expressing human sweet taste receptors and performed Ca2+ imaging on them to analyze the sweetness of recombinant neoculin; sensory tests will also be used to verify the flavoring activity of recombinant neoculin.

Vectors: pEAK10 expression vector, etc.
